Interleukin-17A (IL-17A) is a key cytokine mediating neuroinflammatory processes in mental and neurological disorders, including autism spectrum disorder (ASD). Existing monoclonal antibodies have limited blood-brain barrier permeability, making their application to mental disorders challenging. Currently, the development of low-molecular-weight IL-17A inhibitors is underway. In particular, thiazole-derived inhibitors developed using a scaffold-morphing strategy have demonstrated superior pharmacokinetic profiles while overcoming traditional safety concerns. This review discusses the technical advancements in low-molecular-weight IL-17A inhibitors and their potential for the treatment of mental disorders.
